I want to collect 'Manager' + 'Assistant'

Not sure quite what you're after, but at a guess you could try:
=DCount("*";"az";"(rtba = 'Manager' OR rtba='Assistant') And mort = 'Permanent'")

replace the semicolon with comma.

=DCount("*", "az", "(rtba = 'Manager' OR rtba='Assistant') And mort = 'Permanent'")

I tried it I did not see any error, its working here.
Check maybe your keyboard, change it to english not arabic.
